Brechin are struggling for numbers and will have to promote some youth players to make up their squad.
Gerry McLaughlan and Charlie King are out with long term injuries and Callum Booth has reached the end of his loan.
Annan Athletic manager Harry Cairney: "There is a big prize awaiting us in the next round as we could have our first ever meeting with neighbours Queen of the South at a senior level.
"We played well at Glebe Park and could have won the game up there."
Brechin manager Jim Weir said: "We are a wee bit short of numbers at the moment which is disappointing for such a finely balanced cup-tie.
"We need to play better than we did in the first game otherwise there will no Fourth Round Spot for us.
"We have only managed one game recently but scored five in that one so I am hoping we can carry on from where we left off."
